Optimization of Medium and Culture Condition for Ubiquinone-10 Production Using Agrobacterium tumefaciens ATCC4452

The cultural conditions for bacterial ubiquinone-10 production by Agrobacterium tumefaciens ATCC4452 were optimised by conventional fermentation methodology to minimize the viscosity of the broth and increase the yield of the ubiquinone-10.  A study of complex interactions of oxygen supply and fermentation medium was carried out for improvement of ubiquinone-10 contents and concentration.  An optimized medium composition was analyzed in CSL (corn steep liquor) and sucrose.  This strain produced a 20 mg/g-cell contents of ubiquinone and the concentration of culture broth was 30 mg/L.  The high specific growth rate was obtained by increasing oxygen supply (accomplished by regulation of aeration and agitation rate), while the contents of ubiquinone-10 reduced gradually according to increased oxygen supply rate.  The viscosity of fermentation broth was related to an initial pH and a sugar concentration.  At the high range about pH 8.0 a viscosity was increased but the yield of ubiquinone-10 was not affected.
