Production of Tetraacetylphytosphingosine(TAPS) by a Mutant of Phichia ciferrii ATCC 14091

Experiments were carried out to develop a mutant strain from Pichia ciferrii ATCC 14091 and investigate the effect of sodium acetate on the production of tetraacetylphytosphingosine (TAPS). A high-TAPS-producing mutant was selected by staining with sudan black B and colony shape after the treatment of UV and NTG. Because liposoluble dye such as Sudan black B stained lipid globules in whole cells, we selected high-lipid producing mutants in comparative staining. Among high-lipid producing mutants, we selected a high-TAPS-producing mutant with the rugose mat type. Among the organic acids tested, sodium acetate increased the production of TAPS in a 2.5L jar fermenter culture. In the basal medium containing 4 g/L sodium acetate, TAPS production was 2.4 times greater than that in the same medium without sodium acetate. When sodium acetate was added to the fermentation medium initially, the cell growth was inhibited by sodium acetate; however, the final TAPS concentration was higher than that in the same medium without sodium acetate. As a result, it was suggested that sodium acetate was a stimulator of TAPS production by Pichia ciferrii.
